The signal: model credibility is becoming more explicit

The ISPE programme includes risk-based model credibility. FDA's drug-development AI page identifies its January 2025 credibility guidance as draft. These sources support a discussion about context of use; they do not establish that a particular BioMedAna model is accepted for a regulatory or manufacturing decision. [1, 2]

Our practical starting point is a sentence: this model informs this decision, for this process and operating range, with these inputs, under this review arrangement. If the team cannot complete that sentence, a broad accuracy claim is difficult to interpret. A model can be useful for one task and unsuitable for an adjacent one.

02 / PERSPECTIVE

A digital twin has an operating role as well as a mathematical model

Pharma 4.0 is broader than simulation. A useful twin connects process knowledge and scientific models to the people who own a decision, the evidence they can inspect and the quality processes governing action. A technically elaborate model that nobody is authorized or trained to use has not completed that operating change. [4]

MSAT defines the scientific question; manufacturing and QC establish relevant execution and measurement context; the model owner maintains applicability evidence; QA assesses the controlled use where relevant. Leadership supplies review capacity and change ownership. These are part of the twin deployment, not tasks to leave until after the software demonstration.

03 / PERSPECTIVE

A hypothetical scale-up question needs a bounded claim

Imagine a team comparing two vessels for a CHO process. It wants to examine whether oxygen transfer could constrain the planned operating scenario. A mechanistic calculation might help investigate that question. It would not, on its own, establish the resulting titer, glycosylation profile or overall comparability of the manufacturing process.

Separate the engineering calculation, biological relationship and decision role. Document which quantities are measured, which are estimated and which remain unrepresented. A useful twin can reveal a specific risk without pretending to predict every outcome. The relevant acceptance question is whether its evidence supports the proposed use, not whether its interface resembles the physical vessel.

04 / PERSPECTIVE

Keep calibration and evaluation distinct

Calibration uses observations to estimate or adjust model behavior. Evaluation asks how well the resulting model performs on relevant evidence not used to tune it. The separation should reflect the actual deployment question. Randomly splitting highly related time points can provide a less demanding test than holding out independent runs or a later operating period.

Preserve the split rationale and any limits of the available data. If the dataset is too small for the desired independent test, describe the model as awaiting that evidence. Reference priors and synthetic exercises can support development and numerical checking; they do not become process-specific empirical validation merely because the software executes successfully.

05 / PERSPECTIVE

Evaluate the output that changes the decision

A low average prediction error may be unhelpful if the model misses the boundary that determines the next action. Define the decision-sensitive region before selecting metrics. For a risk screen, the team may care about missed constraints, excessive false alerts and how uncertainty changes the review. Those are different from visual agreement across a whole trajectory.

Use uncertainty deliberately. An interval is useful when its meaning and evaluation are clear, not simply when it makes a plot look cautious. Ask whether the model identifies unsupported operating regions and whether a user can distinguish a supported estimate from extrapolation. An appropriate response may be to request another measurement or decline to quantify the scenario.

06 / PERSPECTIVE

Transfer is a new evidence question

A new cell line, feed strategy, analytical method or equipment configuration can change what the model's inputs and outputs mean. Treat transfer as an impact assessment with explicit dependencies. A calculation remaining numerically stable does not establish that the scientific relationship remains adequate for the new process.

We would prepare a transfer record containing the changed conditions, retained assumptions, new uncertainties and proposed evidence. The decision could be continued limited use, recalibration, further evaluation or a restricted operating range. Avoid treating all changes as harmless or assuming every change requires the same work. The justification should follow the scientific consequence.

07 / PERSPECTIVE

Connect predictions to an operating boundary

Prediction, recommended action and autonomous control are different intended uses. A model evaluated for an offline development discussion is not automatically ready to change a manufacturing setpoint. FDA's process-validation guidance is useful background for the lifecycle of drug manufacturing evidence; this article does not propose replacing process qualification with model output. [3]

Retain model version, data provenance, parameterization, units, calculation results and reviewer interpretation together. When an agent explains the result, the explanation should not add certainty that the calculation lacks. A reviewer must be able to identify what would invalidate the recommendation and which additional observations could resolve the uncertainty.

08 / PERSPECTIVE

Match credibility to the model's influence on the decision

The presence of physics, chemistry or machine learning does not settle credibility. Scientific models already take many forms, including mechanistic simulations, kinetic models and hybrid approaches. The practical question is whether the selected method produces useful evidence for this particular decision.

Add two questions to the model record: how much does the decision depend on this output, and what could happen if the decision is wrong? An exploratory comparison, an investigation recommendation and a controller changing a setpoint have different consequences and evidence needs. Define the quantity being estimated, the conditions where it applies and the acceptance criterion before evaluating performance.

BioMedAna should be judged through an executed task: retained project inputs, stated method and parameters, reproducible outputs, appropriate independent evaluation, limitations and a reviewable user decision. An agent's explanation can make the task accessible. The underlying computation and its evidence must still be inspectable.

A general reaction-kinetics engine is not automatically a qualified stability model, and an engineering calculation is not automatically a validated scale-up recommendation. Each application must earn its own intended use.

09 / PERSPECTIVE

A practical test for scientific execution beyond the conversation

Ask any platform—including BioMedAna—to execute the same scientific task with retained inputs, not simply explain how it would approach it. The result should connect a computational method to a decision that an MSAT scientist can inspect. A good language interface makes that work easier to request and review; the interface alone does not supply the scientific evidence.

  • Physics: change a vessel or operating input, rerun the identified calculation and inspect units, assumptions and the resulting engineering trade-off.
  • Chemistry: change temperature or dosing in a defined reaction network and inspect the rate assumptions, species trajectories and material/energy balance.
  • Hybrid AI: compare the mechanistic baseline with the learned correction using the retained evaluation runs; report where the correction helps and where it is not retained.
  • Operating model: ask a qualified receiving user to challenge the result, document its limits and route the next action through the established process.
10 / PERSPECTIVE

What BioMedAna implements beneath the AI interface

BioMedAna brings together a scientific computation layer and AI-assisted coordination. Its physics-based engineering engine calculates quantities such as impeller power per volume, tip speed, superficial gas velocity and oxygen-transfer estimates from stated inputs. The chemical engine integrates reaction networks with temperature-dependent rates, stoichiometry and batch or semi-batch material/energy balances. These computations are implemented numerical work, not values inferred from the wording of a prompt.

Its hybrid modelling services combine mechanistic process equations with Gaussian-process residual models. The implemented workflows compare learned corrections against a mechanistic baseline on held-out runs and retain corrections where the specified evaluation supports them. The underlying numerical and machine-learning methods are established computational tools; BioMedAna’s contribution is connecting them to scientific data, model evidence and a reviewable workflow.

The model basis must remain visible. Some engineering reference coefficients and defaults are illustrative; they require suitable process/equipment evidence before supporting a stronger operating claim. A numerical benchmark establishes something different from process-specific validation. Ask for the configuration, data lineage, calibration record, independent evaluation and applicability limits of the actual model being proposed.

QUICK ANSWERS

The questions behind the headline

Is calibration the same as validation?

No. Fitting model behavior and independently evaluating its intended use answer different questions.

Can synthetic tests establish process validity?

They can test software and numerical behavior. They do not establish empirical validity for a customer process.

Does a digital twin replace PPQ?

This article makes no such claim. Model support, qualification and operational authority must be justified separately.

How does BioMedAna differ from a conversational layer alone?

BioMedAna implements engineering calculations, reaction-network kinetics with material/energy balances, and mechanistic/data-driven hybrid models. Agents coordinate those services. The calculation inputs, model basis and evaluation evidence should be inspectable for the actual proposed use.
