The signal: an operating model deserves an operating question

ISPE describes Pharma 4.0 as an operating model for the pharmaceutical setting. Its scope reaches beyond the installation of a new application. For a founder or site leader, that creates a useful starting question: which recurring decision would work differently if the organization could use its information more effectively? [1, 2]

The 2026 Annual Meeting programme brings digital systems, quality, governance and workforce capability into the same conversation. Our reading is that a technology roadmap should connect those responsibilities around a real workflow. The agenda establishes topics and speakers; it does not establish that a particular system improved manufacturing performance. [1]

02 / PERSPECTIVE

The operating model connects resources, information systems, organization and culture

Pharma 4.0 is a change in how a pharmaceutical organization works: how people use resources, how information keeps its scientific meaning, how responsibility crosses functions and how teams learn without weakening the pharmaceutical quality system. ISPE explicitly includes organizational, cultural, process and resource dimensions alongside IT. A deployment that adds an AI interface while leaving those handoffs unresolved has addressed only part of the operating problem. [2]

For a site leader, the first design review should therefore include MSAT, manufacturing, QC, QA and the data/system owner. Agree the receiving decision, who can challenge the evidence, what training and review capacity are needed, and how the resulting action enters existing procedures. The scientific technology matters because it serves that operating arrangement.

Pharma 4.0 joins people, process, culture, resources and scientific information around an owned decision. BioMedAna contributes connected data, physics and chemistry computation, hybrid models and AI-assisted coordination to that operating model.THE BIOMEDANA LENS

Our take: define the unit of value before the architecture

Consider a hypothetical biopharma team preparing a weekly process review. The scientist can find trend data, but needs a colleague to explain a material change and another team to reconcile analytical results. The meeting is delayed because the supporting records describe different versions of the same run. A new dashboard might display that disagreement faster without resolving it.

We would define the first deliverable as a reviewable decision packet for a named review. It should state the process question, data cut-off, unresolved issues, accountable reviewer and next action. That is a more useful unit of value than an unspecified ambition to connect the whole site. It also gives the implementation team a reason to stop adding features.

04 / PERSPECTIVE

Map four kinds of work, not four software purchases

The first map should show who performs the scientific work, where the evidence resides, how the decision passes between roles and what behavior the team needs to support. Our suggested implementation map translates the operating-model discussion into tasks; it is not an alternative official ISPE maturity model. A diagram is useful only when a receiving team can act on it. [2]

For the process review, the scientist defines comparability, the data owner maintains identities, the quality partner defines applicable review expectations and the operational owner manages the action. One person may hold several responsibilities in a small company. The responsibilities still need to be explicit, especially when a decision moves to an external manufacturer or partner.

05 / PERSPECTIVE

Start with the quality question and follow the control strategy

A Pharma 4.0 workflow should connect a product-quality objective to the work that protects it. For a hypothetical biologics process, begin with the relevant quality attributes and the process or material factors that could affect them. Identify the measurements, methods, people and controls that make those relationships interpretable. The team must establish which attributes and parameters are critical for this product; a dashboard cannot assign that status by itself. [4]

For example, consider a hypothetical purification review where aggregate content has been identified as a CQA for that product. A pooling comparison needs fraction and sample identities, the relevant assay version, recovery calculations and the approved operating instructions. A model can help compare scenarios only within its supported scope. MSAT evaluates the scientific trade-off, QC owns method interpretation and the authorized manufacturing and quality roles retain control of execution and any required change. A higher predicted recovery cannot overrule the quality constraint.

A scientific model then has a specific job. It might compare engineering conditions or help test a process hypothesis. Its output joins the evidence reviewed by MSAT and Quality, with any consequential action entering the established control path. The learning can inform monitoring, investigation or a controlled change. This connects scientific computation to the pharmaceutical quality system rather than treating the computation as the endpoint.

The operating outcome can include more reliable supply or lower resource use, provided the team measures it within defined quality constraints. A shorter preparation time is useful; it is only one part of the value.

06 / PERSPECTIVE

Build an evidence contract for the handoff

An evidence contract is our term for an agreed minimum record. For this workflow it could contain batch and material identities, time alignment, method versions, source links, known exclusions and reviewer interpretation. It should separate a measured observation from a derived metric and from a hypothesis. The contract belongs to the decision, rather than to the current software vendor.

Test it with a colleague who did not prepare the analysis. Ask them to explain which observations support the proposed action and which uncertainty might change it. If they must reconstruct the history through messages, the handoff remains incomplete. Capture the missing context once, close to the work that produces it, rather than creating another retrospective documentation exercise.

07 / PERSPECTIVE

Treat maturity as capability demonstrated under stress

Connectivity during an ideal demonstration is a weak expansion criterion. Introduce a late result, a disputed sample identity or a temporary source outage. Observe whether the packet shows the limitation and whether the named owner can decide how to proceed. A mature workflow makes uncertainty visible at the point of use.

Avoid averaging away a critical gap. Excellent visualization cannot compensate for unreliable sample matching in a decision that depends on those samples. We would retain a readiness profile with blocking issues, dependencies and owners. Improvements should remove a specific obstacle to the next decision rather than merely increase a single headline score.

08 / PERSPECTIVE

Measure the work that disappears and the work that moves

Record preparation time, clarification requests, reviewer effort and time to an agreed action before introducing the workflow. Repeat the measurement for comparable reviews. Include the work required to maintain mappings, resolve exceptions and review machine-generated material. A saving for one role may otherwise become an uncounted burden for another.

Do not translate time released directly into a financial return without a utilization assumption. Capacity has value when it can be redeployed to a defined task or avoids a real constraint. The first management discussion should describe what changed, how consistently it changed and what still required expert intervention. That is a stronger basis for expansion than usage alone.

09 / PERSPECTIVE

How BioMedAna joins the operating model to scientific computation

BioMedAna connects contextualized process data, scientific computation and AI-assisted work. Its computational layer includes physics-based engineering calculations, a chemical reaction-kinetics engine and hybrid models combining mechanistic equations with learned corrections. Agents can coordinate evidence retrieval, invoke an appropriate analytical service and prepare the result for review; the numerical answer has an identifiable computational basis.

For the operating model, the important connection is the handoff: which input records were used, which calculation or model version ran, what assumptions apply and who owns the next decision. A scale-up discussion, for example, can inspect engineering quantities such as power per volume, gas velocity and oxygen-transfer estimates rather than relying on a fluent description of scale-up. Process-specific evidence determines which estimates are fit for the intended use.

QUICK ANSWERS

The questions behind the headline

What does Pharma 4.0 include beyond IT?

Resources, information, organization, process and culture, built on an established pharmaceutical quality system. Scientific tools must fit the people and operating decisions they support.

Where should a small biotech start?

Choose a repeated handoff with an accountable owner and accessible evidence. Avoid connecting systems without a defined decision.

What is the first useful metric?

Measure the end-to-end effort and elapsed time for a comparable review, including clarification and rework.
